Sporidesmin was capable of producing rapid and reversible swelling of liver mitochondria. The requirement for phosphate appeared to be related to the ability of this anion to alter the conformation of the mitochondrial membrane rather than to its ability to penetrate the membrane. Sporidesmin also stimulated 'state 4' respiration of well-coupled mitochondria, but was without effect on sonicated or Triton X-100 treated preparations.
