Lactogenic hormone from fresh bovine pituitaries purified by the method of Cole & Li was acetoacetylated for 2.5 h at pH 8.4 using a 100:1 molar ratio of diketene and protein. The procedure caused complete inactivation of the hormone. Partial removal of acetoacetyl groups restored 40% of the activity, and complete removal of the block by hydroxylamine hydrochloride treatment (Marzotto et al. Biochimica et Biophysica Acta (1968) 154, 450) restored 60% of the activity.
