Cholesterol esterification and lipids in plasma and liver from newborn and young guinea pigs raised on milk and non-milk diet

Of 6 groups of albino guineapigs, group 1 was of newborn, unfed guineapigs killed shortly after birth; group 2, guineapigs 3 days old receiving maternal milk; group 3, 3 days old and fed from birth on a low-fat, high-carbohydrate adult diet; group 4, guineapigs 3 weeks old receiving the group 3 diet with an adult male guineapig for company; group 5, guineapigs 3 weeks old brought up with their mother and receiving milk in addition to the group 3 diet; group 6, adult males 7 to 14 weeks old, on the group 3 diet. Blood and liver samples were taken at death. Liver tissue was homogenised and centrifuged to provide a liver cytoplasmic extract. Free and total cholesterol, triglycerides, free glycerol, protein, lecithin cholesterol acyltransferase (LCAT) and acyl CoA cholesterol acyltransferase (ACAT) were estimated in plasma and liver.Free and total plasma cholesterol were high in newborn guineapigs but plasma cholesteryl ester formation was low. Plasma triglyceride and glycerol were higher than in the adult. Liver cholesterol concentrations were comparable to adult values but liver cholesterol esterification activity and triglyceride were high. The 3-day-old guineapigs on milk had plasma cholesterol 2.5 times as much as in the newborn. Cholesterol ester production increased greatly during the first 3 days of life. Plasma triglycerides doubled but glycerol remained constant. Liver cholesterol also increased greatly and the amount present as the ester was much greater than in the newborn. Liver cholesterol esterification increased but triglycerides decreased. In 3-week-old guineapigs on the normal diet plasma lipids had decreased to normal adult values. The rate of cholesterol esterification had increased but the net formation of cholesteryl esters had decreased. All the liver lipid and cholesterol values were comparable to those of adults. Guineapigs 3 days old on non-milk diet had higher plasma cholesterol, triglyceride and glycerol and lower cholesterol esterification than milk-fed animals. Liver cholesterol concentrations were lower in group 3 but higher than in newborns. Cholesterol esterification rate was also lower but glycerol and triglycerides were not affected. Group 4 had much lower cholesterol than group 3 animals, but the rate of cholesterol esterification was greater. Plasma triglycerides and glycerol were significantly lower than in group 3. Liver cholesterol fell to adult values and cholesterol esterification rate and triglyceride decreased significantly compared to group 3 animals.
