The F2 toxin of Fusarium species can be rapidly assayed in animal feeds by two methods: either (A) the feed (50 g) is dried, finely ground, defatted by shaking with petroleum ether (150 ml) and kieselguhr (5 g), then the washed flour extracted with methylene chloride or chloroform for 4 hours in a Sokhlet apparatus, the extract being dried on Na2SO4; or (B) the finely ground feed (50 g) is homogenized with kieselguhr (5 g) in 100 ml chloroform-water (9:1) for 5 minutes, washed with chloroform (2X20 ml), and the combined chloroform fractions reduced to 20 ml before drying on Na2SO4. In both cases the resulting toxin extract is separated by thin-layer chromatography, with chloroform-ethanol (96:4) as solvent, and measured by its fluorescence at 360 nm. Recovery rates of 88-92 and 77 83% were obtained by methods A and B, respectively, with maize samples containing 2 ppm of F2 toxin and rates of 80-88 and 42-54% with samples containing 50 ppm toxin.
