The A (albino) and B (brown) mycelial phenotypes of H. capsulatum cannot be differentiated morphologically in their yeast phases. However, on brain heart infusion agar + cystine + 0.001% nile blue A (NBA) incubated at 37 deg C in the dark, type A yeast colonies remain white to light tan while type B yeast colonies become bright blue in 3-5 days. On modified Sabouraud agar + 0.004% bromo cresol purple (BCP) at 37 deg C in the dark, only type A yeasts turn the medium bright yellow in 24-48 h.
