Oxytocin activity measurements in blood plasma of cows during milking

, : Oxytocin activity measurements in blood plasma of cows during milking. Endokrynologia Polska 25(2): 83-93

10 Polish Black-and-White Lowland cows were milked on 2 consecutive evenings, 1-3, 6-8, 13-15, and 20-22 wk after calving, with an 'Impuls' M 901/1 individual quarter milker working at 380-400 mm Hg vacuum and 45 pulsations/min. (i) on the 2nd evening, milking was by the customary method; but (ii) on the 1st, udder massage and fore-milking were omitted. Blood was taken from a cannulated jugular vein 5 min before udder massage (washing), during massage and fore-milking (omitted on 1st day), during cluster application, at 1-min intervals during milking, and 5 and 10 min after completion of milking. Mean values with SEM are tabulated for oxytocin contents of the blood plasma samples [see DSA 27, 3486 & 28, 1516 for method of determination]. Values before milking ranged from 0.08 to 5.1 mu U/ml for (i) and from 0.9 to 1.4 mu U/ml for (ii). During milking, there was a significant (P<0.01) brief increase in oxytocin activity, more marked (but not significantly so) in (i). Highest values were attained after 1 min milking in the 1st-3rd wk after calving [277 plus or minus 120 mu U/ml in (i) and 80 plus or minus 14 mu U/ml in (ii)]. Max. activity declined progressively with stage of lactation. (ii) had no effect on milk yield but prolonged milking time in comparison with (i).
