Studies on the production of milk clotting enzymes by aerobic sporeformers. III. Purification and some properties of the enzyme

, : Studies on the production of milk clotting enzymes by aerobic sporeformers. III. Purification and some properties of the enzyme. Egyptian Journal of Dairy Science 2(1): 45-54

The enzyme was recovered from the Bacillus megaterium culture filtrate by (NH4)2SO4 fractionation (30-60% saturation), and was partially purified by gel filtration on Sephadex G-150. Throughout the process, the enzyme was purified to approx. 40-fold from the crude preparation, with 12% recovery of the original activity. The enzyme was most stable at pH 5-7 but lost its activity quite rapidly below pH 5 and above pH 7.
