Male NMRI mice were given a stock diet to appetite or were starved for 65 h before they were killed. Cells isolated from pancreatic islets were incubated in the presence of glucose 16.7, glucose 16.7 and theophylline 10 or L-leucine 10 mmol/litre. In experiments on glucose-stimulated insulin biosynthesis L-[3,4,5-3H] leucine served as radioactive precursor and L-[2,3-3H] phenylalanine was used as tracer in experiments on leucine-stimulated biosynthesis. There was no difference between islets from fed or starved mice as regards the incorporation rate of leucine into proinsulin-insulin at glucose 16.7 mmol. There was a higher rate of incorporation into islet proteins precipitable in trichloroacetic acid after starvation. When theophylline was added to the islets of starved mice protein synthesis became more normal so that the proinsulin-insulin fraction of the total protein synthesis was as great as in the islets of fed mice. The effect of theophylline on the islets of fed mice was less prominent. Starvation affected neither the leucine-stimulated incorporation of phenylalanine into proinsulin-insulin nor the ratio of radioactivity incorporated into the proinsulin-insulin pool.
