, : Isolation and properties of Acheta accessory gland polysomes. Journal of Insect Physiology 21(4): 889-901

Polysomes were prepared from male accessory glands of Acheta [domesticus (L.)] and factors found important in ensuring the maximum yield included homogenisation at a large excess of buffer to tissue (40:1), the use of 200 mM ammonium chloride as a nuclease inhibitor, the addition of either sodium deoxycholate or Triton X-100 to the buffer before homogenisation and the use of the Dounce homogeniser. The occurrence of cold-induced 'run-off' of polysomes was confirmed by direct measurement. The polysomes reassembled within 15-30 min after the return of the insects to higher temperatures, with restoration of endogenous protein synthetic capacity to control levels; the ribosomes appeared to retain an elevated capacity for poly(U)-directed protein synthesis.
