Adaptation of a Queensland isolate of avian reticuloendotheliosis virus to chick kidney and chick embryo fibroblast cell cultures is described. A fluorescent antibody technique was developed. Serum samples for the first incubation period were diluted 1/20 in phosphate buffered saline, pH 7.2. The second incubation utilized a purified rabbit IgG prepared against chicken IgG and conjugated with fluorescein isothiocyanate. Coverslips carrying cell cultures were mounted in veronal-buffered glycerol-saline, pH 8.6, and examined by dark-field fluorescence microscopy. Two viruses isolated from batches of Marek's disease vaccine were shown to be avian reticuloendotheliosis virus. A retrospective survey of 586 commercial poultry serum samples collected in 1973 to 1975, before the use of vaccines known to be contaminated, revealed antibody to REV in 2 of 14 breeder-layer flocks in New South Wales, and in one of 30 broiler flocks and a closed turkey flock in Queensland. It is concluded that REV occurs as a natural infection in Australian poultry flocks.
