In experiments to assess whether diurnal changes in thyroid 131I metabolism are paralleled by variations in endogenous iodine, uptake of iodide by the thyroid gland was estimated by a double isotope technique. The thyroid:serum iodine ratio and the rate of 131I and 127I secretion from the thyroid were estimated in rats on low-iodine diets for 20 days. The accumulation of iodine by the thyroid was prevented by KClO4 and organic iodine formation was blocked by 6-propylthiouracil.
