P:O ratio and respiratory control of potato under mitochondria decreased with increasing amounts of glutaraldehyde, rate of succinate oxidation being reduced most rapidly in state 3 and 4. The observed uncoupling was due solely to inhibition of state 3, for mitochondria fixed in glutaraldehyde retained their ability to develop protein gradients. Glutaraldehyde increased the affinity of succinoxidase for the inhibitor, antimycin A; this was confirmed by a fluorescence study.
