A light-sensitive, conditional mutant of maize (Z. mays L.) is described. Under conditions which lead to maximum expression of the mutation, the mesophyll chloroplasts are characterized by high chlorophyll a/b ratios, and the virtual absence of light-harvesting chlorophyll a/b-protein (CPII). The proportion of P700 chlorophyll a-protein (CPI) remains relatively unchanged. Loss of chlorophyll a/b-protein is reflected in a concomittant loss of both associated polypeptides with MW of 23 and 25 kilodaltons. The mutant chloroplasts are characterized by a loss of extensive grana structure. The average number of thylakoids per granum is reduced from 4.8-1.2. Expression of the mutation is controlled by light intensity over the range 40 .mu.einsteins m-2s-1 (400-700 nm) to 475 .mu.einsteins m-2s-1. At lower light intensities the total chlorophyll content is reduced to approximately 60% of the wild-type, but pigment composition and ultrastructure appear normal. Intermediate light intensities produce intermediate characteristics. Expression of the mutation does not appear to be significantly affected by light quality or temperature. Further studies of this mutation and other alleles known at the same locus could prove useful in understanding both the genetic control and biochemical mechanisms involved in plastid differentiation.
