A method for isolating oil palm protoplasts

, : A method for isolating oil palm protoplasts. Phyton, Argentina 40(2): 169-178

Oil palm stem and rachis sections were placed in increasing mannitol solutions (5, 9 and 12%) for 2 h at a time, resulting in gradual cell plasmolysis. This was followed by 18 h maceration in 3% Cellulase Onozuka SS + 1.5% Macerozyme + 0.02% Pectinase Fluka in 12% mannitol at 24 deg C in the dark. Young and adult rachis yielded many protoplasts, young leaves yielded a few, but no protoplasts were obtained from adult leaves.
