Results of trials showed that both ammonium oxalate and HCl could be used for the maceration of tissues of young, fully developed and old leaves of potatoes and barley, but HCl was better for determining chloroplast size and number of chloroplasts/cell. A method for the maceration of leaf tissue with these compounds is described together with information on the duration of treatment depending on leaf age.
