A method for rapid multiplication of virus-free potatoes by tuber production in vitro

, : A method for rapid multiplication of virus-free potatoes by tuber production in vitro. Mededelingen van de Faculteit Landbouwwetenschappen Rijksuniversiteit Gent 46: 1031-1042

Tubers were allowed to germinate in the dark at 20 deg C and then exposed for c. 10 days to c. 2000 lux. The explants were disinfected with ethanol, rinsed, plunged for 30 min in 5% calcium hypochlorite with a few drops of Tween 80 and rinsed again 2-3 times. The apical bud and the base were removed and the segments were cultured in glass tubes with various media at 20 deg C, with a light period of 12 h, at 1000-3000 lux. Rapid tuberization was favoured by a temp. of 24 deg.
