Hepatocellular cytoskeleton is the target of phalloidin, a toxic affecting electively actin filaments. Cytoskeleton alterations have been studied in an experimental model of intrahepatic cholestasis induced in the rat by intraperitoneal injections of phalloidin. An increased pericanalicular deposition of actin, without modification of prekeratin immunostaining, has been shown by immunofluorescence. Immunogold staining with an anti-actin monoclonal antibody has been used to characterize the impaired relationships between canalicular structures, pericanalicular cell-junctions and abnormal actin bundles at the ultrastructural level. Our data provide immunomorphological evidence that cytoskeleton rearrangement in phalloidin induced cholestasis involves actin but not cytokeratin filaments.
