Protoplast formation and DNA-mediated transformation of Fusarium culmorum to hygromycin B resistance

This work involved firstly optimizing the protoplast yields for Fusarium culmorum 159026, then setting up a system for DNA-mediated transformation. Higher protoplast yields and more rapid regeneration were obtained when the organic stabilizers sucrose and sorbitol were used rather than NH-4Cl. Successful transformation was achieved using the plasmids pAN7-1 and pHRC containing the Escherichia coli hygromycin B resistance gene (hygromycin B phosphotransferase, hpt) fused to Aspergillus nidulans expression signals.
