Bacterial artificial chromosomes (BACs) overcome many of the disadvantages of yeast artificial chromosomes (YACs), such as instability and chimaerism of YAC clones. The BAC system uses an F factor-based vector; DNA can be cloned with high efficiency, easily manipulated and stably maintained in Escherichia coli. BAC libraries have recently been constructed for Arabidopsis thaliana, sorghum and rice.
