ISSR (inter simple sequence repeats) as genetic markers in Noctuids (Lepidoptera)

, : ISSR (inter simple sequence repeats) as genetic markers in Noctuids (Lepidoptera). Hereditas Lund 136(3): 251-253

Intra- and interspecific variations among noctuid populations (Lycophotia porphyrea, Diarsia brunnea, Xestia baja, X. c-nigrum, X. ditrapezium, and X. rhomboidea) from natural or exploited forests in France were determined by intersimple sequence repeat-polymerase chain reaction (ISSR-PCR). Several primers were employed to evaluate whether informative genomic fingerprints could be generated. Best results were obtained with the (CA)10 primer, whereas (CTGT)4, (GC)10, (TCC)5, (CT)8, (AG)12, (GATA)4, (CTG)5 failed to produce polymorphic PCR products. (GACA)4, (CGAT)4, and (CT)4(CA)5 showed some variability, but were inferior to (CA)10. The DNA profiles, which differed substantially among species, suggesting that ISSR-PCR efficiently differentiated species with similar morphology.
