It has been previously demonstrated that barley (Hordeum vulgare) microspores at the mid-to-late uninucleate stage display strong nucleolytic activity. In this work, a nuclease was partially purified from diffusates of barley (cv. Arda) microspores by using concanavalin A as ligand. DNase in-gel assay showed that the chromatographic preparation contain a single 34-kDa protein with nucleolytic activity.
