Leaf extracts from Trifolium repens and Phaseolus vulgaris contained several acidic low-molecular weight peptides, the fractions of which were obtained among the most acidic amino acids in an automatic amino acid analyzer and purified further with ion-exchange chromatography. Some of these peptides were originally N-acetylated, but the acetyl group could be hydrolyzed away with 2 mol/l HCI. The concentrations of peptides were about 50 nmol/kg wet (fresh) weight, while those of the adjacent amino acids were about 500 nmol/kg and that of aspartic acid about 1 mmol/kg. Hydrolytic, enzymatic and mass spectrometric studies showed the presence of at least three sequences: Aspartyl-glutamate, glutamyl-aspartate and N-acetylaspartyl-glutamate.
