A review of pharmacological studies is presented on natural inhibitors of proteinase (hirudin) and multivalent inhibitors (aprotinin) in an examination of inhibitors which would block enzymes for participating in human enzyme action regulation. The enzyme-substrate complex, its reactions and fixation are described. The role of inhibitors in hypercoagulation, thrombosis, hyperfibrinolysis, hemorrhaging, shock, inflammation and immune complex disorders is discussed.
