The effects of two antigonadal compounds, danazol and the GnRH agonist buserelin, on the endometrial gland were investigated by studying 13 patients having endometriosis. Six of them received 400 mg/day of danazol orally for 6 months. Their endometriums were biopsied periodically to examine the diameter of the endometrial gland and its mitoses as well as the pseudostratification of nuclei. Out of 19 specimens from the buserelin group, 15 specimens showed pseudostratification of nuclei and 13 showed gland mitoses. In contrast, out of 16 specimens from the danazol group, only 3 had pseudostratification of nuclei and only 2 gland mitoses. Further, when comparing small endometrial glands (.ltoreq. 36 .mu.m in diameter) between two groups, 50 per cent of the specimens from the buserelin group showed the pseudostratification of nuclei and gland mitoses while none of the specimens from the danazol group did. Differences of action mechanism(s) of the two compounds might lead to these results. Our demonstration that the high appearance rate of the pseudostratification of nuclei and gland mitoses even in small endometrial glands from patients taking the GnRH agonist buserelin, may help in determining the use of buserelin in examination of an endometrial histology.
