Development of intercellular junctions in the lining epithelium of fetal lamb lungs was examined by light microscopy and EM. At 39 days' gestation, tight junctions (TJ) consist of 3.1 .+-. 1.6 (SD)-5.8 .+-. 2.0 rows of particles on P [protoplasmic]-face ridges and in complementary E[endoplasmic]-face grooves. By 69 days, mirror-image replicas showed junctional strands to be structurally continuous, and functional studies indicate they are tight. From 76 days onwards the TJ consist of 4.5 .+-. 2.0-7.5 .+-. 2.5 smooth, P-face strands and particle-free E-face grooves. Gap junctions are associated with TJ from 58-120 days of gestation. Desmosomes, present throughout gestation, disappear at birth.
