Effect of prostaglandin e 1 alpha adrenergic blocker and beta adrenergic blocker on the hemodynamics of spontaneously hypertensive rats

Spontaneously hypertensive rats (SHR) are widely used as an animal model for human essential hypertension. The etiology and maintenance of hypertension in SHR have been studied but there are few reports on the hemodynamics of SHR. Prophylactic approaches to hypertension in SHR were studied. Long-term administration of propranolol was effective for the prevention of hypertensive diseases in SHR. The effect of propranolol, PG[prostaglandin]E1 and phenoxybenzamine on the hemodynamics of SHR were studied. Anesthesia was induced by light ether; gallamine (2 mg/kg) and .alpha.-chloralose (40 mg/kg) were added. The trachea was intubated and respiration was maintained artificially. The chest was opened by median sternotomy and an electromagnetic flow probe was attached at the root of the aorta for the measurements of cardiac output (CO). Blood pressure (BP) was measured from the femoral artery. Systolic BP, diastolic BP, left ventricular pressure (LVP) and total peripheral resistance (TPR) were higher in SHR than in normotensive Wistar rats. Heart rate showed a tendency to increase in SHR. Hypertension in SHR was largely a result of increased peripheral resistance. Injection of PGE1 into the aortic arch decreased BP and LVP in both SHR and normotensive Wistar rats. PGE1 acted on the peripheral vascular bed and the heart. Injection of propranolol into the jugular vein decreased CO and heart rate but increased BP and TPR in SHR. Injection of phenoxybenzamine into the jugular vein decreased BP and TPR but increased CO in SHR. There was little observable change in normotensive Wistar rats. Apparently sympathetic tone is accelerated in SHR and there is hypersensitivity to .alpha.- and .beta.-blockers. The elevation of BP by propranolol in SHR may be a species specific reaction.
