Electroencephalogram with epidural electrodes revealed spike discharges about 15 min after topical application of .delta.-guanidinovaleric acid (DGVA), that has one more carbon in its chain than amidino-GABA (.gamma.-guanidinobutanoic acid), on pia mater of rats. Spike discharges induced by DGVA were completely suppressed within 10 min by the supplementation of (3R)-(.sbd.)-4-amino-3-hydroxybutanoic acid (L-GABOB) (1 mM) on pia mater. In rats given L-GABOB topically prior to or together with DGVA, spike discharges were not induced by DGVA application. Not only phenobarbital (PB) (2 mg/kg, i.m.) but also diazepam (DZ) (10 mg/kg, i.p.), applied 20 min after the DGVA application, showed no suppressing effects on spike discharges. DGVA could induce spike discharges by topical application 20 min after the injection of PB or DZ. While L-GABOB showed a remarkable suppressive effect on the spike discharges induced by DGVA, two anticonvulsants, which are known to correlate with the GABA-receptor, had no effect on spike discharges induced by DGVA. These findings suggest that DGVA might act directly on the GABA-receptor and might be a specific GABA antagonist.
