In situ estimation of 4C nuclear DNA content in eleven strains of Pisum sativum in two organs, the root and the shoot, reveals a consistent picture. Intraspecific variation is not very marked. The amount of DNA in the shoot appears to be slightly lower than that of the root in most of the strains. The range of variation of DNA is much lower in the shoot than in the roots. The organwise variation in the DNA content is associated with amplification during differentiation. The DNA content in the root meristem does not show any correlation with chromosome size. The difference in total chromosome length in the strains has been suggested to be due to differential condensation and spiralization, and the variation in total chromosome volume principally due to protein components in addition to DNA content.
