More than 700 haploid plants were produced by induced androgenesis in anther cultures from 3 regenerants and 13 hybrids. Their diploidization was achieved by callus cultures of leaves, leaf pedicels and stem pith parenchyma of young haploid plants. More than 80 homozygous lines were produced, which demonstrated their much higher homozygosity, as compared to the standard cv. 'Harmanliiska Basma 163', in the breeding trials at the experimental field Vranya of the Institute of Genetics, Sofia and in the experimental field Markovo of the Tobacco Institute, Plovdiv (Bulgaria). Most of the lines had considerably higher yields and tobacco qualities than the parental cultivars from which the initial hybrids were developed. Some of the homozygous lines are immune to Peronospora tabacina. Anther and tissue culture can be successfully used in shortening the breeding process for developing high yielding and disease resistant isogenic tobacco lines.
