Inhibitory effects of alpha 1 anti trypsin derived asialo oligo saccharides on the liver uptake of the asialo glyco protein

Oligosaccharides were prepared from the glycopeptides derived from human plasma .alpha.1-antitrypsin (.alpha.1-AT) by alkaline-borohydride cleavage of asparagine-oligosaccharide bond. After i.v. injection into rats, the asialooligosaccharides and asialoglycopeptides, but not the undesialylated materials, were rapidly taken up by liver. Competitive inhibition of liver uptake was shown by the injection of labeled asialo-.alpha.1-AT with either asialooligosaccharides or asialoglycopeptides, while the fully sialylated derivatives did not inhibit the uptake. Periodate oxidation experiment with the asialooligosaccharide demonstrated that the integrity of the exposed galactose residue is required for recognition and uptake by the liver.
