Isolation of membrane bound immunoglobulin g from a human lymphoid cell line

A human lymphoid cell line (Seraphina) with surface located IgG was used for the isolation of membrane bound IgG by Nonidet P-40 extraction of 125I-radiolabeled cells. The isolated IgG was purified and partially characterized by gel filtration and SDS-PAGE [sodium dodecyl sulfate-polyacrylimide gel electrophoresis]. The membrane bound IgG was found to belong to the lambda type. The MW of the membrane bound IgG (106 daltons) and the ability of 2-mercaptoethanol to split the aggregated IgG into components with a MW of 150,000 daltons indicates that the IgG of Seraphina cells might be linked by disulfide bonds into a polymeric structure containing either IgG or IgG linked to some other membrane components with the same MW as IgG. Seraphina cells are also endowed with a significant activity of IgG secretion.
