Lack of physostigmine diazepam antagonism

Prompted by reports of physostigmine reversal of diazepam-induced depression, an attempt was made to document this effect in animal experiments. Physostigmine, an acetylcholinesterase inhibitor, has been used successfully in treating overdosage from drugs with known anticholinergic properties such as atropine, amantadine, clozapine and phenothiazine derivatives. Its ability to reverse the central depression produced by diazepam cannot be explained on the basis of cholinergic blockade. Observations of gross behavior in mice and results from rotarod test showed that physostigmine given i.p. in doses of 0.03 to 0.125 mg kg-1 failed to show a modification of the state of sedation produced by diazepam given i.p. in doses of 2-16 mg .cntdot. kg-1. Measurement of the interaction on motor activity even showed significant potentiation of diazepam-induced sedation by physostigmine. In the light of animal studies, the use of physostigmine in the treatment of diazepam toxicity remains questionable.
