Upon exposure of rats to polychlorinated biphenyls (PCB), vitamin A storage decreased and lipid peroxide formation increased in the liver. The study attempted to determine if activated lipid peroxidation was responsible for the reduction of hepatic vitamin A content in the rats given PCB. The involvement of lipid peroxidation and microsomal cytochrome P-450 was ascertained regarding the reduction of vitamin A in the liver of rats given PCB diets. When fed diets containing PCB (0, 0.01, 0.05%) for 10 days, there were dose-related increases in lipid peroxide formation and cytochrome P-450 content and decreases in vitamin A content. Whether .alpha.-tocopherol supplementation suppressed lipid peroxide formation and reduction of vitamin A content in the liver of rats given PCB was also studied. .alpha.-Tocopherol added excessively to the PCB diet abolished the enhancing effect of PCB on lipid peroxide formation; vitamin A content remained at reduced levels, in spite of significant elevation of hepatic vitamin E content. Lipid peroxidation was not directly responsible for the reduction of vitamin A in the liver of rats given PCB.
