Optimal concentrations of pyruvate were determined for measuring the LDH[lactate dehydrogenase] activity, using constant conditions of buffer, NADH-concentrations and pH; a higher substrate concentration in the determination of LDH-activity in the seminal plasma of bull, stallion and rabbit than normally required for human blood serum was found. In seminal plasma of the boar, optimal pyruvate concentration is at approximately 0.6 nM. In seminal plasma of bull, stallion and rabbit a pyruvate concentration of at least 1 mM is recommended. Using lower concentrations, a considerable decrease of activity must be expected.
