P. serratus lactate dehydrogenase (L-lactate: NAD+ oxidoreductase, EC 1.1.1.27) was purified over 900-fold with an overall yield of 10-15%. Gradient polyacrylamide gel electrophoresis yielded 2 bands, both exhibiting LDH activity. SDS [sodium dodecyl sulfate] gel electrophoresis yielded 1 single protein band corresponding to a monomer MW of 36,000. The MW of the tetramer was 140,000. The purified enzyme exhibits non-Michaelian kinetic behavior and displays negative cooperativity. The Km values are 0.24 mM and 16 mM, respectively, for pyruvate and lactate and are independent of temperature in a 10.degree.-30.degree. C range. The enzyme is inhibited by 3-acetyl-pyridine NAD in a non-competitive way. The kinetic properties of the enzyme are discussed with respect to the possibility of conformational changes.
