During the maturation of human tissue, the lactate dehydrogenase (LDH, EC isoenzyme pattern changes from an intermediate pattern with LDH-3 predominating to a pattern with an increased activity of LDH-1 or LDH-5. In this study the LDH isoenzyme pattern is determined in different human lymphocytic cell populations as a parameter of their cell maturation. Thymocytes show an intermediate pattern with a high LDH-3 activity, which confirms the immaturity of the thymocytes. Normal peripheral T [thymus-derived] lymphocytes contain less LDH-3, concomitant with an increased activity in LDH-1 compared to the thymocytes. T lymphoblasts also contain less intermediate LDH-3 than thymocytes, but exhibit a concomitant increase in the activity of LDH-5. This pattern suggests a more anerobic metabolism, which might be an adaptation of the lymphoblasts to less aerobic culture conditions. The significance of the higher LDH-3 activity (comparable to the thymocytes) and the lower LDH-1 activity of the B [bone marrow-derived] lymphocytes with regard to the T lymphocytes is discussed. This study points to the interest of the determination of the LDH isoenzyme pattern of the lymphocytic cells participating in an immunologic response.
