The effect of hydrocortisone on cholesterol biosynthesis in the rat liver was studied in relation to the period of hormonal action, with 14C-acetate and 3H-mevalonate being used as precursors. The experiments were carried out on Wistar male rats distributed into 3 groups. The animals of the 1st group were injected with hydrocortisone in a dose of 5 mg/kg body wt. The biosynthesis of cholesterol was examined after 5 h. The animals of the 2nd and 3rd groups received hydrocortisone in the same dose for 5 days. Two hours before sacrifice the animals of different series were given i.p. injections of 14C-acetate and 3H-mevalonate in a dose of 10 .mu.Ci/100 g body wt. The animals were sacrificed 2 h after administering the precursors. The liver was removed and perfused. Neutral lipids and phospholipids were detected by TLC in silica gel. The number of lipids was measured by the phosphovanilin technique. Radioactivity of the samples was estimated by a liquid scintillation spectrometer. Specific radioactivity of free cholesterol was calculated. Administration of hydrocortisone for 5 days resulted in the decreased cholesterol biosynthesis in the liver. The same effect was produced by administering the hormone for 10 days. At the same time a single dose of hydrocortisone stimulated the synthesis of free cholesterol following 5 h. Administration of hydrocortisone for 5 days in conjunction with 14C-mevalonate as precursor entailed a 2-fold increase in cholesterol biosynthesis in the liver of the experimental animals as compared with control. Prolonged administration of hydrocortisone inhibits cholesterol biosynthesis from the acetate and increases cholesterol formation from mevalonic acid. Hydrocortisone exerts a different effect on cholesterol biosynthesis depending on the time of administration. Hydrocortisone-induced inhibition of cholesterol biosynthesis in the rat liver is accounted for by the inhibition of .beta.-hydroxy-.beta.-methylglutaryl CoA reductase activity produced by the hormone.
