A new technique for obtaining axenic plantlets to investigate histological aspects of host/pathogen relationship is described. Such plantlets are grown in test tubes on an agar medium supplemented with mineral nutriments and covered with fiber glass paper. Compared with other techniques used for plant symbiosis investigations, the proposed technique has certain advantages: absolute axeny for all plant organs; no secondary contamination during the storage and handling of the test tubes; the host plant is the sole source of organic elements available to the pathogen; simple and quick sampling of plant parts for observation. It provides an in vitro system which is simple and easy and in which all parameters can be controlled both for the pathogen and the host plant.
