Valine 2a transfer RNA .**GRAPHIC**. from baker's yeast [Saccharomyces cerevisiae] was degraded with pancreatic RNase, and the fragments obtained were separated by DEAE-cellulose chromatography in 7 M urea at pH 7.4. Rechromatography of some fractions was performed on Dowex 1 .times. 4 (superfine) column or TLC on cellulose. Two dinucleotides, 6 trinucleotides, 1 trinucleotide tetraphosphate, 3 tetranucleotides, 1 pentanucleotide and 1 hexanucleotide were found. Seven minor nucleotides were present: m5C, .psi., m2G, m22G, mIA, hU, T and N, the chemical structure of N being unknown. The methods of enzymatic degradation, separation and characterization of nucleosides and nucleotides by TLC are described in detail.
