The DNA-actinomycin D complex prepared in vitro (using calf thymus DNA) and administered i.v. to mice had a higher toxicity level than actinomycin alone. Actinomycin D and its complex with DNA had an immunodepressive activity (hemolysins and hemagglutinins) which correlated with their toxocity. The complex had an immunodepressive effect when used simultaneously with or after the antigen administration. The use of the same dose 5 days before the immunization did not induce any suppression of antibody production. Accumulation of 14C-actinomycin D bound to the DNA carrier in the spleen, intestine and kidneys proceeded slower than that of the antibiotic alone, while its excretion rate was higher. Pure actinomycin was reutilized in the spleen. No such effect was observed with the use of the antibiotic in complex with DNA.
