Lymphocytes isolated from human blood were stimulated at day 0 by phytohemagglutinin. They were subjected to gamma irradiation at various times with respect to the initial stimulation. The incorporation of bromodeoxyuridine took place on the 3rd day. The cells were then labeled by the anti-bromodeoxypridine-fluorescein isothiocyanate antibody after hydrolysis with HCT 4N. A sample of non-activated but irradiated lymphocytes were labeled with propidium iodine which binds solely on double-stranded DNA. The preliminary results justified the precise consideration of the delay between irradiation and the initial activation for a more complete exploration of cellular radioprotection.
