Much is not known about the intracellular signal transduction pathways induced by ultraviolet (UV) irradiation. Our study demonstrated that ultraviolet C(UVC, 254nm) irradiation increased 2.2 kb c-fos transcripts in quiescent NIH3T3 cells in a manner similar to tumor-promoting agent, 12-O-tetradecanoyl phorbol-13-acetate (TPA) and serum growth factors. c-fos inductions by UVC irradiation and serum were markedly attenuated by protein kinase C (PKC) inhibition. In contrast, TPA inducibility of c-fos was completely inhibited in the same condition. These results suggest that PKC is activated during the cellular response to UVC irradiation and other protein kinases may also participate in signal transduction following UVC irradiation.
