1,2-Dimethylhydrazine certainly induced colon cancer in mice and its histology demonstrated moderate to well differentiated adenocarcinoma. We divided carcinogenesis into three steps based on CHANG's classification. Using light and electron microscopic lectin histochemistry, we investigated changes in the carbohydrate chain of glycoprotein and the results obtained were as follows: 1) No differences in the staining pattern was detected among atypical crypts, early neoplastic lesion, and neoplasm. 2) In contrast to normal colonic epithelia, decreased staining of PNA and RCA-1 in the Golgi apparatus and positive labeling of UEA-1 on the apical plasmalemma were found. Positive staining of DBA on the basolateral plasmalemma was also observed in the neoplasm. 3) Tumor cells sometimes possessed mucin granules stained with WGA, DBA, and UEA-1 lectins. These findings suggest that galactosyltransferase activity decreases and fucosyltransferase activity increases in the tumor cells.
