The fraction P from WEHI-3B cell conditioned medium has neither granulocyte-macrophage colony stimulating activity nor burst promoting activity itself but augments large colony formation of macrophages in semi-solid culture of bone marrow cells in the presence of colony stimulating factor. To clarify the presence of fraction P responsive cells in granulocyte-macrophage progenitor cells, we studied the effect of fraction P on DNA-synthesis by murine marrow granulocyte-macrophage colony forming units (CFU-GM) in short-term liquid culture. The addition of fraction P to the liquid culture medium resulted in maintenance of the number of CFU-GM assayed after 6-24h of incubation. The proliferation of CFU-GM before and after exposure to fraction P was studied by hydroxyurea suicide. It was found that fraction P enhances the percentage of CFU-GM synthesizing DNA after 6-24h of incubation in liquid medium. Fraction P could not support the survival of the interleukin 3 dependent FDCP-2 cell line. These results suggested that DNA synthesis-inducing factor in fraction P was different from interleukin 3.
