We found previously that mammalian bile contains a plasminogen activator, bilokinase, and that this activator is continuously secreted by the liver into the biliary tract. However, little is yet known about how the bilokinase activity in the bile changes in relation to liver disorders. We therefore measured several parameters of coagulation and fibrinolysis including the bilokinase activity in experimental liver cirrhosis in rats. The cirrhotic rats were recognized as being at the initial stage of cirrhosis involving inflammation: there was an elevated plasma fibrinogen level, prolonged euglobulin clot lysis time (ELT) and shortened prothrombin time (PT) as well as a shortened activated partial thrombin time (APTT). However, in the cirrhotic liver, the production of plasminogen activator was augmented. The bilokinase activity of the cirrhotic rats was significantly increased as compared to that of normal rats, demonstrating that hepatic fibrinolysis exerts a direct effect on biliary fibrinolysis.
