The response of the isolated rabbit heart to ischemia and reperfusion with and without manganese superoxide dismutase (Mn-SOD) treatment in summer and winter has been compared. Isolated, buffer-perfused hearts were subjected to 1 hour of global ischemia followed by 1 hour of reperfusion, and the percentage recovery of developed tension (relative to preischemia levels) was measured. The coronary effluent was assayed for creatine kinase release. Mn-SOD, when administered, was included in both preischemic and postischemic buffers. The results show that the injury sustained from a given insult exhibits wide seasonal variation. Mn-SOD provided significant protection, regardless of season. Although the cause of this variation is unclear, it may lead to erroneous conclusions regarding drug efficacy, unless controls and treatment groups are contemporary.
