The subcellular distribution of exogenously administered horseradish peroxidase (HRP) in pancreatic acinar cells was investigated after induction of pancreatitis in rats by infusion of a high dose of caerulein. In addition, membrane binding of five lectins with distinct sugar specificities was determined. In rats with pancreatitis, HRP was rapidly sequestered into lysosomal autophagic vacuoles. In contrast, no HRP reaction product was detected in secretory granules and vacuoles known to contain digestive enzymes. Membranes of secretory vacuoles showed a low degree of lectin binding in contrast to a strong membrane labeling of autophagic vacuoles. Binding of lectins to both types of vacuoles suggests that at least a part of the outer membrane derives from the Golgi elements. In contrast, only autophagic vacuoles seem to be involved in endocytotic processes via the basolateral plasma membrane.
