A high-performance liquid chromatographic (HPLC) method was developed for the simultaneous determination of three protoberberine type and one aporphine type alkaloids in Phellodendron amurense Rupr. Berberine, palmatine, jatrorrhizine and magnoflorine were separated by HPLC on an ODS-column with acetonitrile-1/15 M sodium dihydrogen phosphate-sodium lauryl sulfate (45:55:0.2, v/v/w) as the mobile phase with detection at 270 nm. The quantitatives of the four alkaloids in the trunks, branches, leaves, leafstalks, fruits (seeds) and roots of P. amurense Rupr. were determined by the present method. The berberine content of the bark on trunks slightly increases with the decrease in the height of sampling point, while the palmatine, jatriorrhizine and magnoflorine concentrations concomitantly increase with the increase in the height of sampling point. It was confirmed that berberine is contained in the leaves and leafstalks of P. amurense Rupr.
