Children with growth hormone (GH) deficiency commonly have poor sexual development, ranging from micropenis in the newborn period to delayed puberty. GH has been shown to have a synergistic effect with adrenocorticotropic hormone on adrenal steroid synthesis. These observations suggest that GH may play a role in the testicular steroid secretion. We studied sex prepubertal males, ages 8-15 years, with isolated GH deficiency. Serum testosterone levels were measured before, during and 24 hours after the intramuscular administration of human chorionic gonadotropins (hCG) 2,000 IU for three consecutive days, prior to and after three months of methionyl-GH replacement (0.1 mg/kg three times per week). The testosterone response to the administration of hCG was significantly increased (p < 0.003) after three months of GH replacement. The results suggest a synergistic effect of GH on luteinizing hormone (LH) action on the biosynthesis of testosterone in the testes. Whether GH exerts its effect by the modulation of LH receptors, stimulation of testicular somatomedin-C or increasing the activity of testicular hyroxylases remains to be determined.
