Immunohistochemical localization and distribution of cholecystokinin octapeptide in porcine, rat and guinea pig intestinal tract

Cholecystokinin (CCK), a peptide with multiple molecular forms and physiological functions, is widely distributed in both the nervous system and the endocrine cells of digestive tract. In this study, the localization and distribution of cholecystokinin octapeptide (CCK-8) in the porcine, rat and guinea pig intestinal tract were investigated by ABC immunohistochemical technique, and the possible coexistence of CCK-8 with serotonin was also observed by using adjacent section double immunolabelling method. It was found that the CCK-8 immunoreactive (CCK-8-IR) cells were maidy localized in the lower parts of intestinal glands, while only a few could be observed on the epithelium of villi. In the aspect of segmental distribution. the CCK-8-IR cells could be seen in the mucosa of the whole intestinal tract from duodenum to colon in pig, while in rat and guinea pig, the CCK-8-IR cells were only seen in the mucosa from duodenum to ileum. But, in the aspect of quantity, the density of CCK-8-IR cells showed the highest number only in duodenum. Nearby the adjacent sections of the duodenum, the results obtained by double immunolabelling method proved the coexistence of CCK-8 and serotonin in the intestinal tracts of all three animals.
